No.  You send a message after the package has been confirmed as uploaded.
There is no reason to wait.  Just make sure your release announce language
mentions that it will be uploaded to a mirror soon.

I think it is better to do these things as soon as possible, otherwise
experience has shown that people forget.

I can't remember if this was one of those setup.hints which inexplicably
contained a '@ mined' line, but if it does please make sure that it
is removed.  It is not needed.
